I recently noticed a leak in the 4 or 5 inch main drain to my house. The pipe is inconveniently located behind my furnace and water heater so i had to go through the drywall in my utility room to access it. What i have found is that it only leaks when mass quantities of water are flushed through it. If i just lather the joints with ABS glue will that be sufficient?

you may NOT attempt a 'rube goldberg' repair on a sanitary waste line
but
go ahead and waste your time and effort - glue is cheap

As above, but make sure the water isn't coming down from another location and just following the pipe. You already said that you can't see it well, so there could be a different issue.

I had a plumber look at it and he says it could cost as much as 250$ to replace the trouble section. I want to attempt the repair myself but am not sure how to get the new piece of pipe in place because there is no flex in the line. Do they make flexible ABS for these situations?

This fix is NOT a recommended repair but likely will fix the leak the ABS pipe is not under pressure and leaks can be made to stop leaking MAYBE This is not a professional recommendation and best only for the home owner until it is fixed properly A soldering gun can soften the pipe if you are creative to seal a leak so guess you get the drift

When an ignition source is removed from pvc, flames go out. When they are removed from ABS, it still keeps burning, rolling toxic black smoke all the while. Don't try to melt ABS with anything except the solvent designed for the purpose.

Vertical PVC piping will NOT, repeat NOT self extinguish !
Try this OUTDOORS for yourself:
Clamp a length of 2" vertically ~ light the bottom ~ VOILA, TURBO-TORCH !
INTERNALLY BURNS JUST LIKE A TORCH
This is why PVC was/is NOT allowed in high-rise structures as drainage piping stacks or venting between floors.
REGARDLESS OF CODE VERTICAL PVC STACKS OR ANY VENTING WILL BOTH BURN AND SPREAD FLAMES TO OTHER FLOORS (not to even mention the toxic gasses released).
